ABSTRACT

OBJECTIVE@#Numerous studies have demonstrated the close relationship between chronic stress and blood pressure (BP). Hypertensive subjects exhibit exaggerated reactions to stress, especially higher BP. The mechanisms by which stress affects pre-existing hypertension still need to be explored. Danzhi Xiaoyao Powder (DP), a historical traditional Chinese medicine formula, is a promising treatment for BP control in hypertensive patients under stress. The present study investigated the metabolomic disruption caused by chronic stress and the treatment effect and mechanism of DP.@*METHODS@#Spontaneously hypertensive rats (SHRs) were subjected to chronic restraint stress (CRS) for 4 weeks. BP was measured via the tail-cuff method, and anxiety-like behavior was quantified using the elevated-plus-maze test. Meanwhile, DP was administered intragastrically, and its effects were observed. Global metabolomic analysis was performed using ultra-high-performance liquid chromatography-quadrupole time-of-flight mass spectrometry, followed by multivariate statistical analysis to detect differential metabolites and pathways.@*RESULTS@#DP alleviated the CRS-induced increase in BP and anxiety-like behavior. Systematic metabolic differences were found among the three study groups. A total of 29 differential plasma metabolites were identified in both positive- and negative-ion modes. These metabolites were involved in triglyceride metabolism, amino acid (phenylalanine, tryptophan, and glycine) metabolism, and steroid hormone pathways.@*CONCLUSION@#These findings expose the metabolomic disturbances induced by chronic stress in SHRs and suggest an innovative treatment for this disorder.

ABSTRACT

Objective:To explore the mechanism of modified Xiaoyao Powder on inflammatory response of rats with syndrome of stagnation of liver qi and spleen deficiency of experimental autoimmune thyroiditis (EAT) from the perspective of differentiation of microrna 326 (miR326) regulating Th17 cell.Methods:48 rats were randomly divided into normal group (12 rats) and model group (36 rats) respectively and they were immunized twice a week with high iodine water combined with subcutaneous injection of thyroglobulin. From the fifth to eighth weeks, 36 rats were immunized once a week. From the fifth week, the model group with liver depression and spleen deficiency syndrome of Traditional Chinese Medicine was reproduced with chronic restraint stress, excessive fatigue and eating incoherence methods. The modelrats were randomly divided into model group, Xiaoyao Powder group and Jinshuibao group. Rats in Xiaoyao Powder group were gavaged with 13.63 g/(kg·d) Xiaoyao Powder modified granule suspension, and rats in Jinshuibao group were gavaged with 477 mg/(kg·d) Jinshuibao suspension, twice a day, for 8 weeks.The levels of serum FT3, FT4, TSH, TGAb and TPOAb were detected by ELISA; the expression of miR326, IL-17 mRNA, IL-4 mRNA and IFN-γ mRNA were detected by PCR. The expression of Ets-1 protein in thyroid tissue was detected by Wes method, and the proportion of CD4 + IFNγ + T cells, CD4 + IL-4 + T cells and CD4 + IL-17 + T cells were detected by flow cytometry, HE staining was used to detect the pathological manifestations of thyroid tissue in each group. Results:Compared with the model group, the serum TSH [(3 328.88±724.45) pg/ml vs. (1 900.25±203.91) pg/ml] in Xiaoyao Powder group increased ( P<0.01), TGAb [(63.60±9.01) IU/ml vs. (96.19±10.74) IU/ml] and TPOAb [(6.84±1.45) IU/ml vs. (11.62±2.06) IU/ml] decreased ( P<0.01), and the expression of miR326 (3.57±0.57 vs. 7.63±0.90),IL-17 mRNA (6.71±0.97 vs. 13.02±1.18) significantly decreased ( P<0.01), the expression of Ets-1 (0.71±0.40 vs. 0.39±0.02) significantly increased ( P<0.01), the ratio of CD4 +IFN-γ + T cell [(13.10±2.23)% vs. (20.7±2.07)%], CD4 +IL-17 + T cell ratio [(18.90±1.31)% vs. (25.1±1.03)%] significantly decreased ( P<0.01), and thyroid histopathology changed significantly. Conclusion:Modified Xiaoyao Powder could regulate the expression of target protein Ets-1 upward, inhibit the differentiation of Th17 cells and further reduce the expression of IL-17 mRNA by regulating the expression of mir-326 downward in the thyroid tissue of EAT rats, so as to improve the inflammatory response of rats with liver depression and spleen deficiency.

ABSTRACT

This article is to explore the antidepressant mechanism of Shugan Lipi recipe in regulating tryptophan metabolism,and to find out their common pharmacodynamic substances. UPLC-Q-TOF-MS technology was used to establish fingerprints of Shugan Lipi recipe,and 124 components were identified. The depressed mouse model was replicated by triple-one multiple stress method. Chaihu Shugan Powder,Sini Powder and Xiaoyao Powder were administered in groups to observe the changes in body weight and behavior of the mice. The results showed that compared with the model group,the body weight,sucrose preference percentage and autonomous activity behavior of each administration group were improved. Among them,the effect of Chaihu Shugan Powder was better than that of Sini Powder and Xiaoyao Powder. LC-MS/MS method was used to determine the contents of 5-hydroxytryptamine( 5-HT),kynurenine( KYN) and tryptophan( TPP) in blood,liver,brain,colon and other tissues,as well as TDO enzyme activity in liver. Western blot and RT-PCR were used to detect the protein and gene expression of TDO enzyme,respectively. It was found that the three prescriptions increased the ratio of 5-HT/KYN in different degrees,decreased the ratio of KYN/TRP in liver,colon and brain,and decreased the expression level and activity of TDO enzyme in liver. The order of their ability to regulate tryptophan metabolism was Chaihu Shugan Powder>Sini Powder>Xiaoyao Powder. In addition,the correlation between the chromatographic peaks in the fingerprints of Shugan Lipi recipes and the pharmacodynamic indexes of tryptophan metabolism was analyzed by the grey relation analysis. The grey relation analysis found that the chemical components with the highest correlation with tryptophan metabolism were mainly from Paeoniae Radix Alba,Citri Reticulatae Pericarpium,Aurantii Fructus Immaturus and Aurantii Fructus. UPLC-Q-TOF-MS was used to analyze the migration components in the plasma of mice after administration of Shugan Lipi recipe,and to verify the common pharmacodynamic substances of Shugan Lipi recipe. The migration of these detected components in plasma was studied,and a total of 18 prototype components and 36 metabolites were identified. Therefore,it was believed that Chaihu Shugan Powder,Sini Powder and Xiaoyao Powder could play an antidepressant role by reducing the expression of TDO enzyme in the liver and regulating the metabolism of tryptophan.The components contained in Paeoniae Radix Alba,Citri Reticulatae Pericarpium,Aurantii Fructus Immaturus and Aurantii Fructus were the common pharmacodynamic substances of Shugan Lipi recipe,which played an important role in regulating tryptophan metabolism.

ABSTRACT

Objective: To characterize the endogenous metabolites and metabolic changes of feces of chronic unpredictable mild stress (CUMS) rats by 1H-NMR, evaluate the improvement effects of Xiaoyao Powder and investigate the underlying mechanisms. Methods: The depression model was established by CUMS procedure. 1H-NMR coupled with multivariate statistical analysis was employed to reveal the changes of fecal metabolic profiles of CUMS rats and identify potential bio-markers involved in CUMS-induced depression. Based on the potential bio-markers, the relevant metabolic pathways were constructed. Results: A total of 10 metabolites was identified as potential bio-markers in fecal samples for the CUMS model. Compared with the control group, the contents of asparagine, aspartate, lactate and propionic acid in the CUMS rats were significantly increased (P < 0.05, 0.01), while phenylalanine, tyrosine, glutamate, glutamine, alanine and proline were significantly decreased (P < 0.05, 0.01). The administration of Xiaoyao Powder could significantly increase the levels of phenylalanine, tyrosine, glutamate, glutamine and proline, whereas reduced the levels of asparagine, lactate and propionic acid. Compared with the control group, six metabolic pathways were recognized as the most influenced pathways associated with the CUMS-induced depression: (1) aminoacyl-tRNA biosynthesis, (2) alanine, aspartate and glutamate metabolism, (3) arginine and proline metabolism, (4) glutamine and glutamate metabolism, (5) phenylalanine metabolism and (6) pyruvate metabolism. Among them, Xiaoyao Powder significantly mediated abnormalities of five pathways of (2), (3), (4), (5) and (6). Conclusion: It is the first report to investigate the antidepressant-like effects and underlying mechanisms of Xiaoyao Powder from the perspective of fecal metabolites. The current results showed that the anti-depression mechanisms of Xiaoyao Powder might be related to regulating the amino acid metabolism, glucose metabolism and intestinal microbial metabolism. This study provides a solid basis for revealing the anti-depression mechanisms of Xiaoyao Powder comprehensively and deeply.

ABSTRACT

Objective: To study the effects of Xiaoyao Powder (XYP) and its composition drug groups (Shugan group and Jianpi group) on the behavior medicinal effects of depressed rats. In addition, the rough set theory and the model of decision rules were used to analyze the interaction between composition drug groups and their effects on the anti-depression of XYP. This study will provide the research method and theoretical basis for the scientific explanation of the compatibility rule of anti-depression of XYP. Methods: The depression rats model was constructed by Chronic Unpredictable Mild Stress (CUMS). The effects of XYP and its composition drug groups on CUMS rats were investigated in open field test, sucrose preference rate and forced swimming. Afterwards, Rosetta software was used to construct the decision rules model of rough set. The interactions among XYP, Shugan, and Jianpi groups were analyzed according to the decision rules. Results: XYP and its composition drug groups significantly improved the depressive behaviors of CUMS rats. Compared with the model group, XYP and its composition drug groups significantly increased the number of crossings and rearing of the rats in the open field test (P Jianpi group > Shugan group. The order of improving the behaviors in the open field test was Shugan group > Jianpi group > XYP. The order of decreasing the time of forced swimming index was XYP = Shugan group > Jianpi group. The results of decision rules showed that, Shugan and Jianpi groups exhibited certain antidepressant effects when used alone, but they were weaker than that of XYP. Of note, Shugan and Jianpi groups exhibited synergetic effect in a dose-dependent manner: The higher the dose was, the greater the antidepressant effect was. In order to further verify the mutual relationship between Shugan and Jianpi groups on the anti-depression efficacy, according to the decision table of XYP and its individual herbs, attribute reduction of eight herbs in XYP was conducted, obtaining the representative drugs of Shugan and Jianpi groups: Bupleuri Radix and Paeoniae Radix Alba. The results of decision rules were consistent with that of composition drug groups. Conclusions XYP and its composition drug groups can not only improve the core symptoms of depression, including index in open field test, sucrose preference rate and forced swimming index, showing remarkable antidepressant effects. The effects of XYP on these indexes were stronger than that of its composition drug groups, indicating that the composition of the XYP prescription is well-suited and scientifically reasonable. The decision rules based on rough set theory could be used as an approach to scientifically interpret the compatibility laws of TCM prescriptions.

ABSTRACT

This study aimed to investigate the effect of the petroleum ether fraction of Xiaoyaosan (XY-A) in a rat depression model with consideration of an underlying mechanism based on gut microbiota and metabolomics. All procedures involving animal treatment were approved according to the Committee on the Ethics of Animal Experiments of Shanxi University. A rat model was established using the chronic unpredictable mild stress (CUMS) procedure and XY-A and venlafaxine (positive control) were used as intervention drugs. Sequencing of the 16S rRNA gene combined with LC-MS metabolomics was used to investigate the effects of XY-A on gut microbiota and metabolites in CUMS-induced depression, and Pearson correlation analysis was carried out on gut microbiota and metabolites. The results showed that XY-A significantly improved the depression-like behavior of CUMS rats and restored the level of brain-derived neurotrophic factor (BDNF) in the hippocampus. Gut microbiota analysis revealed that XY-A can increase the diversity of microbial species in CUMS rats and significantly restored the relative abundance of intestinal Rothia [Prevotella], with effects on intestinal inflammation and the production of short-chain fatty acids. Cecal content metabolomics identified twenty biomarkers that were altered by depression, whereas administration of XY-A ameliorated the changes in seventeen metabolites, with the most strongly affected metabolic pathways being linoleic acid metabolism, taurine and hypotaurine metabolism, primary bile acid biosynthesis, and arginine and proline metabolism. Correlation analysis further showed that there was a strong relationship between the gut microbiota and the cecal content metabolites. In summary, XY-A may exert antidepressant effects by regulating the composition of the gut microbiota and the metabolites and pathways of the cecum. The results provide a reference for the potential molecular mechanism of antidepressant action of XY-A.

OBJECTIVE@#To study the effect of soothing liver, relieving depression, invigorating spleen and reinforcing blood on reducing delirium in elderly patients with hip fracture.@*METHODS@#From December 2014 to June 2018, 180 elderly patients with hip fracture admitted were divided into treatment group and placebo group according to the order of admission:90 patients in treatment group were treated with Jiawei Xiaoyao Powder(JWXYP), including 32 males and 58 females, with an average age of(72.12±4.92), involving 67 cases of femoral trochanter fractures and 23 cases of femoral neck fractures; 35 cases underwent dynamic hip screw fixation, 31 cases underwent intramedullary fixation and 24 cases underwent artificial hip replacement. In the placebo group, 90 patients were treated with placebo, including 37 males and 53 females, with an average age of(72.91±5.43) years old, involving 69 cases of femoral trochanteric fractures and 21 cases of femoral neck fractures, including 37 cases underwent dynamic hip screw fixation, 30 cases underwent intramedullary fixation and 23 cases underwent artificial hip replacement. The age, sex, injury site, intraoperative bleeding volume, postoperative drainage, operation time, anesthesia time, post-operative pain score, post-operative hemoglobin, post-operative CRP, delirium severity(DRS) score and delirium occurrence were observed and compared between the two groups.@*RESULTS@#All patients were followed up until delirium returned to normal, postoperative delirium was found in 12 cases (13.33%) in the treatment group and in 39 cases(43.33%) in the placebo group, the treatment group was significantly better than the placebo group. The monitoring indexes of the two groups were compared:post-operative pain score(=0.002), post-operative hemoglobin(=0.012), post-operative CRP(=0.042).@*CONCLUSIONS@#JWXYP can relieve liver depression, invigorate spleen and invigorate blood circulation, reduce pain, inflammatory stimulation and supplement blood volume after operation, and significantly reduce the incidence of delirium after operation.

ABSTRACT

Objective: In this paper, herb-component-common target network and protein interaction network were constructed to study the mechanism of Xiaoyao Powder in the treatment of depression and diabetes for exploring the common mechanism of Xiaoyao Powder in treating these two diseases. Methods: TCM database@Taiwan, TCMID, Batman-TCM database and literature mining were used to obtain the components and targets of Xiaoyao Powder, combined with CTD, TTD, PharmGKB, OMIM, Gencards, and Drugbank databases to obtain the targets of Xiaoyao Powder in the treatment of depression and diabetes. The herb-component-common target network was constructed by Cytoscape software. The tool of Metascape was used to perform GO biological processes analysis, Reactome pathway analysis, KEGG pathway analysis, protein interaction networks construction and module analysis on the common targets of Xiaoyao Powder in the treatment of depression and diabetes. BioGPS database and Genecards database were used to analyze the tissue distribution and subcellular distribution of the hub targets, and the tissue-target network and subcellular-target network were constructed by Cytoscape. Protein class was performed on hub pathway targets by DisGeNET database. Results: The results showed that the immune and inflammatory responses, G protein-coupled receptors, insulin and insulin receptors, and monoamine neurotransmitter were involved in the treatment of these two diseases by regulating cAMP signaling pathway, Calcium signaling pathway and PI3K-Akt signaling pathway by Xiaoyao Powder. Conclusion: It provides a direction for further research on Xiaoyao Powder’s common mechanism in treating depression and diabetes. It suggests that the efficacy of Xiaoyao Powder in the treatment of depression and diabetes are mainly involved with the regulation of BDNF-related signaling pathways, the process of G protein-coupled receptors, monoamine neurotransmitter and the insulin and insulin receptors, which will benefit to the further investigation on the declaration of the characteristics of the pharmacological mechanism of Xiaoyao Powder.

ABSTRACT

Using the literature retrieval tool Arrowsmith to explore the possible modern medical mechanisms of Xiaoyao Powder and single herb in the treatment of depression from the theoretical level. The related literature retrieval tool Arrowsmith was used to search and find the associated literature. The results were analyzed and summarized from the single drug, compound of Xiaoyao Powder, and common key terms. Correlation analysis between the composition of Xiaoyao Powder and the mechanism of antidepressant effect by Cytoscape tool. The results showed that different single drugs in Xiaoyao Powder may affect the nerve-endocrine-immune and other key pathways by regulating TLR4, TRPV1, Caspase-3, PPARG, COX-2, NRF2, PI3K, ERK1, MAPK, p38 MAPK, and other targets. Anxiety, restraint stress, spleen deficiency, learning, and memory play an antidepressant role. The main acting site of Xiaoyao Powder anti-depression is in the hippocampus. The antidepression effect of Xiaoyao Powder embodies the characteristics of multi-component, multi-target and multi-pathway of traditional Chinese medicine. Through Arrowsmith search, this study provides a scientific basis for further elucidating the mechanism of antidepression effect of Xiaoyao Powder, and provides research directions and ideas for the research of traditional Chinese medicine compound prescription.

ABSTRACT

Metabolomics, as an important part of systematic biology, is more closely related to phenotype, and has the characteristics of integrity, objectivity, sensitivity and dynamics, which enables it to be rapidly applied and popularized in the research field of traditional Chinese medicine. In this paper, the application of metabolomics in antidepressant pharmacological research, screening of pharmacodynamic markers and development of new antidepressant drugs were summarized. Xiaoyao Powder can achieve the therapeutic effect through the rectifying effect of energy metabolism and intestinal microflora metabolism. Amino acids, sugars, and other substances may be the diagnostic markers of depression or therapeutic markers of Xiaoyao Powder. On the basis of fully exploring the scientific connotation of the antidepressant effect of Xiaoyao Powder, a new antidepressant medicine was created to provide new ideas and explore new approaches for the inheritance and development of the classic formula.

ABSTRACT

Objective: The network pharmacology method was used to predict the active components, targets, and mechanism of Xiaoyao Powder and Kaixin Powder, and to explore the molecular mechanism of depression “same disease with different treatments”. Methods: TCMID, Batman-TCM, and TCM database@Taiwan were used to predict the targets and ingredients of Xiaoyao Powder and Kaixin Powder. The prediction was made via screening out the antidepressive drug targets approved by TTD, PharmGkb, CTD, Genecard, OMIM, DrugBank database, and constructing protein interaction network (PPI). Through the Network Analyzer tool of Cytoscape, the topology of the target was analyzed, the MCODE algorithm of the plug-in ClusterViz was used for cluster analysis, and the DAVID biological molecular function software was used to perform enrichment analysis. Results: The network analysis indicated that two prescriptions may play antidepressant roles through INS, AKT1, TP53, IL6, and CREB1 targets, involving neuroactive ligand-receptor interaction, cAMP signaling pathway, serotonergic synapse, calcium signaling pathway, and cholinergic synapse pathways. The unique antidepressant mechanism for Xiaoyao Powder is that PIK3CA, BDNF, PIK3R1, and NGF target the PI3K-Akt, calcium, and other signal pathways, and CCND1, PRKACA, GCLM, GCLC targeting oxytocin signaling pathway, cholinergic synapses, and other pathways is considered as Kaixin Powder unique antidepressive mechanism. Conclusion: Xiaoyao Powder and Kaixin Powder have similarities and differences in antidepressant targets due to their differences in composition, and the scientific connotation of “same disease with different treatments” is preliminarily clarified by the analysis of the target and the pathway.

Objective: To provide the proper dose specification in pharmacodynamics and clinical application, the composition and dose of Xiaoyao Powder were clarified in Prescription of peaceful benevolent dispensary. Methods: The primary composition and dose of Xiaoyao Powder were verified according to the ancient medical documents, ancient weights, estimate, the results of modern research and so on, so as to confirm the specification conversion standard.Results: According to the weights and estimate in the Song dynasty, the dose of Xiaoyao Powder can be converted as follows: one Liang at that time is equal to 41.3 grams at present. Conclusion: The cognition of the composition and dose of Xiaoyao Powder was obscure and confused for a long time, which made the study of pharmacodynamics quite different.Only by using the primary composition and dose standard can make the results of researches on its medical effects reliable.

ABSTRACT

Depression is a common mental illness that seriously harms to the physical and mental health of human beings. Due to its biological-psychological-social interaction pathogenic feature, researchers hold different opinions for the pathogenesis of depression. Recently, accumulating the clinical and basic researches on the comorbidity of depression and diabetes make the roles of glucose metabolism, especially central glucose metabolism and insulin resistance in the pathogenesis of depression get much more attention. It has been believed that the "liver" and "spleen" are closely related to each other physiologically and pathologically. A large number of studies have confirmed that the core biological connotation of liver-spleen and pathology of traditional Chinese medicine is "emotion-energy network imbalance", which has similarities with the mechanism of glucose metabolism imbalance in depression. Therefore, it is necessary to combine the biological connotation of the liver and spleen with the modern medicine to conduct the further research, which will consequently provide scientific basis for further exploring the pathogenesis of depression and finding complementary and alternative treatment for depression.

ABSTRACT

@#AIM: To observe the clinical efficacy of Xiaoyao Powder combined with tobramycin dexamethasone in treating dry eye syndrome caused by Meibomian gland dysfunction(MGD). <p>METHODS: Totally 134 eyes of 106 dry eye patients with MGD in the First People's Hospital of Wolong District, Nanyang City from November 2015 to November 2017. They were randomly divided into the control group and the observation group, each with 53 cases(67 eyes). The two groups were all given hot compress massage, and other basic treatment. The control group was given tobramycin dexamethasone treatment, while the observation group was treated with Xiaoyao Powder on the basis of the control group. The clinical effect of two groups of patients, the symptoms of dry eye, corneal fluorescence staining(FL), the scale of quality of life for diseases with visual impairment(SQOL DVI), tear film break-up time(BUT)and the Schirmer test(SⅠT)were observed. <p>RESULTS: After treatment, the total effective rate of treatment in the observation group(86.6%)was significantly higher than that of the control group(67.2%). The symptoms of dry eye, corneal FL and SQOL DVI scores were significantly lower than those in the control group, and the values of BUT and SⅠT were significantly higher than those in the control group, the difference was statistically significant(<i>P</i><0.01). During the treatment period, the incidence of intraocular pressure elevation in the observation group was significantly lower than that in the control group, the difference was statistically significant(<i>P</i><0.05). <p>CONCLUSION: The Xiaoyao Powder combined with tobramycin dexamethasone in the treatment of MGD dry eye can significantly improve the curative effect and improve the prognosis of the patients.

ABSTRACT

Objective To investigate the therapeutic effects of modified Xiaoyao Powder combined with abdominal needling for conducting fire back to its source for the treatment of swallowing dysfunction in Parkinson's disease, and to explore a more effective therapy for swallowing dysfunction in Parkinson's disease. Methods Forty-two Parkinson's disease patients suffering from swallowing dysfunction were divided into treatment group and control group, 21 cases in each group. The patients in both groups received routine swallowing function training. In addition to the training, the treatment group was treated with modified Xiaoyao Powder combined with abdominal needling for conducting fire back to its source, and the control group was given conventional western medicine of Domperidone and Levodopa Benserazide Hydrochloride. After treatment for 3 weeks, we compared the dysphagia scores and effective rate of the two groups. Results (1) All of the patients in the treatment group completed the trial, but 2 cases of the control group dropped out. Therefore, a total of 40 cases completed the trial. (2) The dysphagia scores of the two groups were much improved after treatment (P<0.01 compared with those before treatment) , and the improvement of the treatment group was superior to that of the control group (P < 0.05). (3) After treatment for 3 weeks, the total effective rate of the treatment group was 95.24% and that of the control group was 52 . 63%, and the difference was statistically significant between the two groups(P<0.01).(4) Gastrointestinal reaction occurred in 3 cases(one from the treatment group and 2 from the control group) , but there were no other side effects found in both groups. Conclusion Modified Xiaoyao Powder combined with abdominal needling for conducting fire back to its source is more effective for the treatment of swallowing dysfunction in Parkinson's disease than routine western medicine treatment.

ABSTRACT

ObjectiveTo investigate the efficacy of Baihe Xiaoyao powder in patients with chronic hepatitis B (CHB) and depression, and to explore its mechanism of action from the perspectives of endocrine and immune function. MethodsA total of 120 patients with CHB who visited the Outpatient Department of Zhejiang Hospital of Integrated Traditional Chinese and Western Medicine from September 2013 to February 2015 were enrolled, and according to the Hamilton Depression Scale (HAMD) scores, they were divided into CHB-depression group (80 patients) and CHB group (40 patients). The levels of cortisol (Cor) and interleukin-2 (IL-2) and the proportion of natural killer (NK) cells were compared between the two groups. The 80 patients in the CHB-depression group were further divided into control group and treatment group, with 40 patients in each group. The patients in the control group were treated with entecavir for 12 weeks, and those in the treatment group were treated with entecavir and Baihe Xiaoyao powder for 12 weeks. The effects of Baihe Xiaoyao powder on liver function [alanine aminotransferase (ALT) and aspartate aminotransferase (AST)], hepatitis B virus (HBV) DNA, HAMD score, Cor, and immune function (IL-2 and NK cells) were observed in patients with CHB and depression. The t-test was used for comparison of continuous data, the chi-square test was used for categorical data, and correlation analysis was also performed. ResultsCompared with CHB patients without depression, those with depression had a significantly higher level of Cor, a significantly lower level of IL-2, and a significantly lower proportion of NK cells (P<0.01), and Cor level was negatively correlated with IL-2 level (r=-0.786, P<0.01) and the proportion of NK cells (r=-0.681, P<0.01). After the treatment with Baihe Xiaoyao powder for 12 weeks, compared with the control group, the treatment group had significantly lower HAMD score and levels of Cor, ALT, and AST (P<0.01) and significantly higher level of IL-2, proportion of NK cells, and HBV DNA clearance rate (P<0.05). ConclusionThe CHB patients with depression have endocrine disorders, an increased serum level of Cor, a reduced level of IL-2, and a reduced proportion of NK cells. Baihe Xiaoyao powder can improve depressive emotion, reduce the level of Cor, improve the body's immune function and liver function, and increase HBV DNA clearance rate. A combination of Baihe Xiaoyao powder and antiviral therapy can improve the clinical outcome of CHB.

ABSTRACT

Objective To observe the effect of Bacillus coagulans tablets Combined with minus Xiaoyao powder, Nifuratel nystatin soft capsule in treatment of VVC. Methods A total of 108 cases of VVC. patients were randomly divided into 2 groups:observation group and control group. In the observation group (n=54), patients were given vaginal placement of Nifuratel nystatin soft capsule, 1 capsule daily, for 6 days;oral administration of minus Xiaoyao powder, 1 agent daily, for 7 days; and oral administration of Bacillus coagulans tablets, 3 tablets one time, 3 times a day, for 14 days, double dose at first. In control group (54 cases), patients were given vaginal placement of Nifuratel nystatin soft capsule, 1 capsule daily, for 6 days;and oral placement of vitamin C tablet, 1 tablet one time, 3 times a day,for 14 days. The short-term effect and the long-term recurrence of were observed 3-7 days, 1 months, 3 months, 6 months after stopping drug treatment. Results 3 days after stopping drugs, the effective rate in the observation group and the control group was 92.59%and 81.48%, respectively, the difference between the two groups was statistically significant (P<0.05) . 6 months after stopping drugs, the recurrence rate was 9.26% in the observation group, and 20.37% in the control group, the difference was statistically significant (P<0.05) . Conclusion For Vaginal candidiasis, on the basis of antifungal therapy, giving Bacillus coagulans tablets for regulation of intestinal micro ecological environment, and traditional Chinese medicine minus Xiaoyao powder for clearing heat and removing dampness, soothing the liver, can significantly improve the short term and long term curative effects,and reduce the recurrence rate.

Objective] To evaluate the navel therapy of ancient formula for ehloasma of liver qi stagnation syndrome. [Method] The 40 cases take navel therapy of ancient formula, change every 24 hours for one month; make therapeutic effect observation before and after the treatment . [Result] 21 cases were cured, 10 had marked effect,7 had effective,2 had no effect, and the total effective rate was 95%.[Conclusion] Navel therapy of ncient formula has good cure effect on ehloasma.

Objective: This current study focused on the identification of active constituents from Angelica Sinensis Radix in Xiaoyao Powder based on UPLC-PDA-guided isolation technique. Methods: The UPLC-PDA chromatogram of Xiaoyao Powder was compared with that of Angelica Sinensis Radix. The relative retention time of each peak and the Uhraviolet spectra provided by PDA were used in the analyses. The constituents were isolated from Angelica Sinensis Radix under the guidance of UPLC-PDA investigation. The structures of the isolates were elucidated by NMR techniques. The antidepression effect was evaluated on glutamate-induced neurons. Results: Five marker peaks of Xiaoyao Powder fingerprint belonged to Angelica Sinensis Radix and they were determined as coniferyl ferulate (1), E-butylidenephthalide (2), ligustilide (3), Z-butylidenephthalide (4), and 14-acetoxy-12-senecioyloxytetradeca-2E,8E,10E-trien-4,6-diyn-1-ol (5). Compound 5 was isolated from the plants in Umbelliferae for the first time. The treatment with compounds 1, 3, and 4 could protect PC12 and SH-SY5Y cells from glutamate-induced cytotoxicity. Antidepression bioactivity of compound 1 was first investigated. Conclusion: UPLC-PDA-guided isolation technique is confirmed to be a rapid and accurate method to identify the main active constituents from Angelica Sinensis Radix in Xiaoyao Powder.

Objective:To study the effect of Xiaoyao Powder on Alzheimer's disease in hippocampal CA 3βregion of rats PP-2A,GSK-3βexpression.Methods:Rats were randomly divided into four groups , including model group, western medicine group,Chi-nese medicine group were treated with intraperitoneal injection of D-gal and A-β1-42 peptide bilateral hippocampal injection mold-ing, physiological group only with equal volume of sterile saline intraperitoneal and hippocampal injection molding .The model was completed, normal group, model group were perfused with saline , western medicine group and Chinese medicine group were treated with oxiracetam solution and Xiaoyao decoction , four groups of intragastric volume was 0.5 ml/100 g, 1 time a day, continuous 28 d. After intragastric administration of isolated rat brain immediately , packet marking in 4℃4%glutaraldehyde solution of glass container , stored at 4℃.Repair piece, cut from the hippocampus, embedded in paraffin, sliced.Dilution of PP-2A for 1∶200, GSK-3βdilution of 1∶150.Images were analyzed by using Image-pro Plus 5 image analysis system, data were conducted by SPSS11.5 statistical analysis software, the comparison between 4 groups by single factor analysis of variance , between the two two groups was compared by LSD-t test, the test level of α=0.05.Results:The expression of PP-2A positive cell number and the positive area , average optical density, integral optical density index , in Xiaoyao Powder orally intervention compared with the model group increased significantly (P<0.01);GSK-3βabove indices were significantly decreased than that in model group after Xiaoyao Powder after intragastric ad -ministration (P<0.01).Conclusion:Xiaoyao Powder can up regulate the expression of PP-2A and down regulate expression of GSK-3β, may be condensed A-β1-42 peptide induced hyperphosphorylation of Tau has certain inhibition .
